  • 1. Modeling of calcium ions release from non-excitable cells
    • 1.1. Literature survey (past experiments, data analysis and previous theoretical models).
    • 1.2. Design and implementation of computing algorithms; Numerical simulations for testing; Optimizations.
    • 1.3. Large scale numerical simulations for a larger number of calcium-release events; Finding relevant calcium signal properties for different fluorescence indicators; Comparison with existing experimental data. Measurements to be performed in Catania / Italy

    • 1.2. Literature survey on past data and experiments.
    • 2.2. Preliminary measurements on proliferation and viability of Jurkat line cells. Start of a spectrofluorimeter setup.
    • 2.3. Preliminary measurements of intracellular Ca2+ (spectrofluorimetry), collaboration with inst. Catania / Italy.

  • 3. Estimation of implicit parameters of calcium signals in non-excitable cells.
  • 3.1. Extraction of observable calcium parameters from all simulated calcium signals.
  • 3.2. Finding the dependency functions between observable and implicit parameters. Spectrofluorimeter set-up completion. Lab. chemicals, services for cell-culture set-up lab.
  • 3.3. Theoretical calculation of distribution of observable parameters, in relation with a large number of events.
  • 3.4. Comparison between theoretical and experimental observed distributions. Estimation of in-vivo values (average ionic current, release time, etc.).
  • 3.5. FInding a method for estimation of implicit parameters from fluorescence imaging data.
